The invention relates to a process for in-vitro plum tree multiplication and rooting on artificial nutritive media of semisolid consistency containing wheat starch and gelling agent. According to the invention, the plum tree (Prunus domestica) propagation is performed on a modified Murashige & Skoog nutritive medium to which there are added 0.5 mg/l of 6-benzyl-amino purine and 50 g/l of starch as a gelling agent and the rooting takes place on the same starch-gelled medium, without hormones in the composition. The components are admixed into the composition before the autoclave treatment which is performed at 121°C, for 30 min, for sterilization purposes. The explants employed for the in-vitro culture are shoots fragments of 1-2 cm which are inoculated as follows: 5...7/vessel for the multiplication stage and 15/vessel for the rooting stage. The cultures are incubated in a growth chamber with controlled atmosphere of 23...26°C while applying light of about 2400 lux for 16 h/day. The multiplication cycles last for 2...3 months and the rooting cycles last for 1...2 months, high-quality plant material being obtained thereby.
