Provided are cloning and applications of semi-dominant gene qGL3 capable of controlling grain length and grain weight of a rice kernel. In the present application, a major QTL gene qGL3 (SEQ ID NO: 1) which is capable of simultaneously controlling the grain length and grain weight of a rice kernel and a semi-dominant allele qGL3-D (SEQ ID NO: 3) thereof having an epistatic effect are separated from rice and cloned. The two genes are capable of improving the yield and appearance quality of rice. At the same time, two homologous genes qGL3-L1 and qGL3-L2 of the gene in rice are cloned and it is proved that the two homologous genes have a regulation effect on the length of the rice kernel. Thus, the gene qGL3 and the epistatic allele qGL3-D and two homologous genes qGL3-L1 and qGL3-L2 thereof all can be applied to crop genetic modification.
