摘要
PURPOSE: A method for detecting a mutant gene with drug resistance to an HIV drug using restriction fragment mass polymorphism is provided to early diagnose an HIV drug resistant mutation in an HIV-infected patient and to allow the patient to medicate a suitable drug, thereby preventing the patient from missing a treatment period or blocking worsening diseases.;CONSTITUTION: A method for detecting a mutant gene with drug resistance to an HIV drug comprises the steps of: amplifying a polynucleotide template containing a mutation site of a reverse transcriptase gene or a protease gene which exists in an HIV pol gene using a forward primer and a reverse primer; cleaving the amplified polynucleotide using restriction enzyme and generating two or more kinds of single stranded polynucleotide fragments which contain the mutation site and have the limited number of bases; and analyzing the presence of a mutant gene with drug resistance in the reverse transcriptase gene or the protease gene and the mutation kind of the gene by measuring molecular weight of the fragments.;COPYRIGHT KIPO 2013;[Reference numerals] (AA) Codon 215T; (BB) Codon 215Y; (CC) Mass per unit charge, m/z (mass-to-charge ratio)
