PURPOSE: A magnetic nanoparticle coated with silica which is modified by adding hydrophobic groups and functional groups for protein purification and a manufacturing method thereof are provided to effectively isolate and purify protein in a short time. CONSTITUTION: A manufacturing method of a magnetic nanoparticle coated with silica comprises the following steps: dispersing a magnetic nanoparticle in cyclohexane in which oleic acid is added; stirring the magnetic nanoparticle dispersion solution after adding a mixture of poly(oxyethylene) nonylphenyl ether and cyclohexane; while keeping pH within 10-12 by adding a base, obtaining a silica coated magnetic nanoparticle by driving after refluxing the mixture at 15-40 deg. Celsius for 20-24hours by adding tetraethyl orthosilicate; after dispersing the silica coated magnetic nanoparticle in an organic solvent, adding a compound which has more than one functional group which is selected from 3-mercaptopropyl-trimethosysilane and bis(3-triethoxysilylpropl)disulfide and an alkyltriethoxysilane which has an alkyl group with 12-18 carbon atoms in order to increase a temperature up to 100-130 deg. Celsius; refluxing for 6-8hours under high speed stirring; and obtaining a produced reactant by washing and drying.
