Preparing an yeast strain, comprises: providing an initial prototrophic yeast strain; providing at least one integration cassette comprising nucleic acid sequence of an inducible promoter by a substrate of yeast fermentation and framed of two sequences of homologous recombination for inducing the integration of the integration cassette of gamma-glutamylcysteine synthetase gene (GSH1) and/or glutathione synthetase (GSH2) gene; and introducing the integration cassette into the starting yeast strain by transformation. Preparing an yeast strain in which at least a copy of endogenous gamma-glutamylcysteine synthetase gene (GSH1) and/or glutathione synthetase (GSH2) gene are placed under the control of an inducible promoter by a substrate of yeast fermentation, comprises: providing an initial prototrophic yeast strain; providing at least one integration cassette comprising nucleic acid sequence of the inducible promoter by a substrate of yeast fermentation and framed of two sequences of homologous recombination for inducing the integration of the integration cassette of GSH1 or GSH2 gene; and introducing the integration cassette into the starting yeast strain by transformation. Independent claims are included for: (1) the yeast strain obtained by the method; (2) an yeast obtained by culturing the strain; (3) a method-I of producing yeast comprising culturing the strain in a culture medium in aerobic condition; (4) a method-II of producing an yeast extract or an autolyzed yeast extract comprising supplying the yeast and hydrolyzing or autolyzing the yeast; and (5) a method-III of producing glutathione comprising supplying the yeast or yeast extract or autolyzed yeast extract, and extracting glutathione.
