DISTINGUISHING CELLS IN SAMPLE USING INACTIVATING EXTRACELLULAR ENZYME BEFORE RELEASING INTRACELLULAR ENZYME

PROBLEM TO BE SOLVED: To provide a method for detecting the absence or presence of target cells in a liquid sample.;SOLUTION: In a method for detecting the absence or presence of target cells in a liquid sample: (a) (i) the sample comprises an extracellular medium containing an enzyme with a measurable activity, and (ii) the sample is suspected of containing intracellular enzyme target cells with the measurable activity; and (b) the method comprises the steps of (i) treating the liquid sample with a reagent that inactivates the measurable activity in the extracellular medium but does not inactivate the measurable activity in the target cells; (ii) lysing the target cells to release the intracellular enzyme; and (iii) measuring the measurable activity.
