The present invention relates to a reaction mixture L3 for the molecular detection of potato coil virus (PLRV) by means of quantitative RT-PCR, which is characterized in that it comprises a 1.mu.l sample of tested RNA, a reagent comprising a mixture of reverse transcriptase and DNA polymerase enzymes, a mixture of nucleotides, buffer containing magnesium chloride, MgCl.sub.2, sterile distilled water supplementing the reaction mixture to a final volume of 25 .mu.l primers with a specific L-4561F nucleotide sequence: GTT GGT GAA GCG GAT GGT GTC AAA, L-4691R : ATT GTA GCG TCC CGT TCA AGG AGT 130 bp fragment and probe L-4595S: TA CGC AAC GAC AAC ACC TAC CGC CAA, where the probe is labeled with a fluorescent dye 6-carboxyfluorescein (FAM) and used as a quencher tetramethylcarboxyrhodamine (TAMRA).
