A method of determining a vertical distribution and dimensional structure of zooplankton in water reservoir relates to ecology, fish facilities and can be used for evaluating functioning biotic community of aqueous ecosystems aimed at collecting data concerning number, biomass, and space distribution of key hydrocole species. The method includes underwater video filming with a laser illumination and counting organisms observed in the frame using a computer analysis of images in the video file or visually, at that using a narrow frontal laser illumination of the organisms, and all the organisms in the scanned water column are continuously recorded, and the individual depth of location and size of each organism, as well as the vertical distribution of the organisms of each size group are determined, wherein the vertical distribution of organisms of each size group is determined by the formula; ;where xi,j concentration of zooplankton organisms of a j size group in a i layer of z height; ni,j is a number of organisms of a j size group with a d registered vertical position in a z(i-1)≤dz∙i range; h is height and l is width of video camera visibility at a level of laser plane.
