A procedure for chromatography wherein the sample solution is prepared in a solvent or mixture of solvents having elution strength greater than the mobile phase, comprising: equilibrating the stationary phase with a weak mobile phase; injecting the sample solution in the column; elution wherein mobile phase is changed from a weak mobile phase to a strong mobile phase using a gradient (initial gradient); and further elution in either isocratic mode with strong mobile phase or in gradient mode having lesser slope than the initial gradient.
