The insulin analogue or its pharmacologically permissible salt containing two polypeptides forming chain A and chain B is characterised by a general formula 1, where X is the basic amino acid and means lysine or arginine,and R stands for inert amino acid, selected from glycine, alanine, serum or treonine, the amino acid sequence A being selected from SEQ ID No 1 to 4, and the amino acid sequence B being selected from SEQ ID No 5 or SEQ ID No 6, and that the insulin analogue has an isoelectric point of values from 6 to 8. The result also includes prolonged therapeutic composition, use of insulin analogue, dosage regimen and treatment of diabetes mellitus. The solution concerns stable insulin compounds, pharmacologically active, prolonged, flat,the peak glucose concentration during repeated administration and at the same time showed no strong daily fluctuations in glucose concentration, i.e. saw teeth effect.
