The present invention relates to a method of manufacturing magnetic mesoporous silica for strong surface bond with the protein of a bacterial cell wall, and a high efficiency bacteria separation and detection method using the same. The present invention comprises: a step of preparing mesoporous silica; a step of obtaining precursor adsorption silica by putting the prepared mesoporous silica in a precursor solution and then agitating them for 2-3 hours; a step of obtaining magnetic mesoporous silica with pores by reducing the precursor adsorption silica under a hydrogen atmosphere at 450 to 550C for 2 to 3 hours; a step of putting the magnetic mesoporous silica in a transition metal aqueous solution containing transition metal ions and then agitating them for 30-60 minutes; and a step of filtering the agitated mixture and vacuum-drying them at -70 to -80C for 60 to 90 minutes. Accordingly, the present invention can conveniently separate and concentrate bacteria in a short time by using the magnetic mesoporous silica manufactured by an economical method without the existing complex separation step, and can lower detection limit.
