PROBLEM TO BE SOLVED: To improve the speed and specificity of nucleic acid amplification in a method for isothermal nucleic acid amplification using turnback primer (TP).SOLUTION: As shown in Fig. (D1), in a target nucleic acid sequence, a sequence (A) is selected; and as an annealing sequence on the 3'-terminal side of a primer, a sequence (Ac') complementary to the sequence (A) is determined. As shown in Fig.(D2), in the target nucleic acid sequence, a sequence (B) present toward the 5'-terminal side of the sequence (A) is selected, and as a stem loop-forming sequence on the 5'-terminal side of the primer, the same sequence (B') as the sequence (B) is determined. As shown in Fig. (D3), in the target nucleic acid sequence, a sequence (W) between the sequence (A) and the sequence (B) and a sequence (Wc) complementary to the sequence (W) on the 3'-terminal side of the sequence (A) are selected; and as a sequence between the sequences (Ac') and (B') of the primer, the same sequence (W') as the sequence (W) is determined.
