Human skin tissue sampling methods and models to test hypotheses about the mechanisms that cause skin pigmentation, and methods for causing skin pigment levels in ex vivo skin tissue. The method comprises the steps of preparing a first cultured human skin tissue sample, testing the effect of a test agent to cause skin pigmentation, and a transcription profile generated from the treated human skin tissue sample. Comparing the control transcription profile to verifying the hypothesis that the test agent causes skin pigmentation.
