PROBLEM TO BE SOLVED: To accurately detect anaerobic ammonia-oxidizing bacteria.SOLUTION: A sample containing anaerobic ammonia-oxidizing bacteria is stained by lipid staining agent. The stained sample is irradiated with Green excitation light (G excitation) having wavelength 500 to 560 nm to detect red fluorescence near 600 nm. Location of lipid (i.e. bacteria) stained by Nile red is detected by the detected red fluorescence. Further, the stained sample is irradiated with Blue excitation light (B excitation) having wavelength 450 to 490 nm to detect green fluorescence near 500 to 570 nm. Ladderane lipid specifically excreted from anaerobic ammonia-oxidizing bacteria is detected by the detected green fluorescence. Screens obtained by G excitation and B excitation are superposed, anaerobic ammonia-oxidizing bacteria is discriminated from bacteria stained by the lipid staining agent.
