PPROBLEM TO BE SOLVED: To provide a method and a reagent for the separation of nucleic acid from a biospecimen to easily separate long-chain nucleic acids in high yield, concentration and purity and provide a nucleic acid separation kit to use the separation method. PSOLUTION: The method for the separation and purification of nucleic acid from a biospecimen comprises (a) a step to add a biospecimen dissolution reagent containing a buffering agent, a surfactant and a chaotropic agent to a biospecimen to dissolve the specimen, (b) a step to add a nucleic acid precipitation reagent containing one or more kinds of alcohols to the solution prepared by the step (a) to precipitate the nucleic acid, (c) a step to recover the precipitated nucleic acid from the biospecimen solution containing the precipitated nucleic acid and obtained by the step (b), (d) a step to add a nucleic acid cleaning reagent to the precipitated nucleic acid recovered by the step (c) to clean the precipitated nucleic acid, (e) a step to add a nucleic acid dissolution reagent to the precipitated nucleic acid cleaned by the step (d) to dissolve the precipitated nucleic acid, and (f) a step to recover the nucleic acid solution obtained by the step (e).
