The present invention relates to a composition comprising a Vitis vinifera radix extract or a fraction thereof, as an active ingredient, for preventing or treating hepatitis C. More particularly, a Vitis vinifera radix extract, which is extracted with water, alcohol or a mixed solvent thereof, or a fraction of the extract has an excellent effect of selectively inhibiting genomic replication of hepatitis C virus with very low toxicity to interstitial cells. Thus, the composition can be useful for prevention and treatment of hepatitis C.
