The present invention relates to a novel method for measuring in vitro cytotoxicity in earthworm coelomocytes and a composition thereof, and more specifically, to a novel and sensitive method for measuring in vitro cytotoxicity of two types of earthworm Eiseniaandrei and Peryonyxexcavates, and to a composition thereof. To this end, the method comprises the steps of extracting earthworm coelomocytes from an earthworm, exposing the extracted coelomocytes in a copper solution, dyeing the coelomocytes with calcein-AM (calcein-acetoxymethyl ester), and measuring the same by flow cytometric analysis.
