The present invention relates to a novel method for differentiating pancreatic beta-cells using a combination treatment of Pref-1/EGF/insulin in order to increase a proliferation and differentiation efficiency of pancreatic stem cells using combination treatment technique of Pref-1/EGF/insulin. Therefore, the method has advantages of developing a transplant source capable of improving beta-cell loss and hypofunction which is a main disease cause of diabetic and progression thereof, in the case of transferring stem cells to beta-cells using a method for differentiating pancreatic beta-cells of the present invention.
