Disclosed herein are agents that modulate the cis interaction between repulsive guidance molecule A (RGMa) and neogenin or lipid rafts. Modulation with the agent can include blocking cis interaction between RGMa and neogenin and / or destroying lipid rafts. This in turn promotes neuronal survival and axonal growth and / or regeneration. Also disclosed herein are methods for treating disease in a subject in need thereof. The method can include administering the agent to a subject. Further disclosed herein are methods for identifying agents that modulate the cis interaction between RGMa and neogenin.
