The present invention relates to scaffold proteins derived from plant cystatins and nucleic acids encoding them. The scaffold is highly stable and has the ability to form proteins. Scaffolds are particularly well suited for the construction of libraries in, for example, phage display or related systems. The invention also relates to various uses of the scaffold (including therapeutic, diagnostic, environmental and security monitoring, synthetic biology and research uses) and cells and cell cultures expressing the scaffold protein.
