The present invention provides, among other things, methods for quantifying the effectiveness of capping mRNA, in particular mRNA synthesized in vitro. In some embodiments, methods of the present invention include providing a sample of mRNA containing capped and non-capped mRNA, providing a cap-specific binding substance under conditions that allow a complex to be formed between the cap-specific binding substance and the capped mRNA, and quantifying the amount of the complex in comparison control with counting the efficiency of mRNA capping.
