The present invention discloses a method for detection of at least one of genus Burkholderia and Burkholderia pseudomallei species in a sample. The method includes selecting Burkholderia genus -specific and Burkholderia pseudomallei species-specific genes from a gene database, designing one or more set of primers, amplifying the selected genes and internal amplification controls using the one or more set of primers by PCR, optimizing two duplex PCR formats, Format I & II, for amplifying Burkholderia genus -specific region and B. pseudomallei specific region present the sample, wherein including the internal amplification control in the duplex PCR formats to avoid false negative results, and comparing the duplex PCR formats with the monoplex PCR formats for the selected genes to match amplified gene sequences, thereby detecting at least one of the genus Burkholderia and the Burkholderia pseudomallei species in the sample.
