Disclosed is a polynucleotide library comprising a plurality of compositions, wherein the plurality of compositions comprises cDNAs encoding variable regions that belong to the same clonal family, wherein: each composition is present in a separate container; the composition comprises cDNA molecules derived from a single sample, wherein the cDNA molecules are covalently attached to a sample identification region, and the nucleotide sequence of the sample identification region is distinct from the nucleotide sequence of the sample identification region of other compositions present in other separate containers in the library, and wherein the cDNA molecules derived from the single sample comprise: (i) cDNA molecules that encode immunoglobulin heavy chain variable regions and cDNA molecules that encode immunoglobulin light chain variable regions from one or more B cells; or (ii) cDNA molecules that encode T cell receptor alpha chain sequences and cDNA molecules that encode T cell receptor beta chain sequences from one or more T cells.
