Discloses a method of identifying a yeast cell that produces a multi-subunit complex with a greater purity, comprising: (a) providing a panel of yeast cells, said panel comprising at least two yeast cells that each comprise differing numbers of copies of one or more of the genes encoding the subunits of the multi-subunit complex, wherein at least one of said genes encoding the subunits of said multi-subunit complex is not contained in a polycistronic gene; (b) culturing each said yeast cell to express the multi-subunit complex; (c) measuring the purity of the multi-subunit complex produced by each said yeast cell; and (d) identifying the yeast cell that produces a greater purity than another yeast cell in said panel of yeast cells as a yeast cell that produces a multi-subunit complex with a greater purity.
