Provided is a medical radiation positioning film and a method of photographing lesion site, positioning of which is convenient and fast. There is a photo-development fibrous layer (13) in the medical radiation positioning film. When using a C-arm machine to irradiate, a nonradiable structure in the photo-development fibrous layer (13) shows a meshed covering surgical area in a radiation image. Doctors can determine a surgical site and a spatial positional relationship of location membrane markers according to corresponding special marks, so as to achieve a precise operation. Both a first intermediate transparent layer (12) and the photo-development fibrous layer (13), and the photo-development fibrous layer (13) and a second intermediate transparent layer (14) are bonded by a binder therebetween. The photo-development fibrous layer (13) can also be directly printed on one side of the film layer, wherein a photo-development grid is printed in the form of a dotted line, a solid line or a combination of a dotted line and a solid line.
