The present invention relates to a three-dimensional cell culture system having a culture space including holes with predetermined cross-sections and depth, which are arranged at regular intervals in two or more rows and two or more columns, wherein the cell culture system is composed of porous polymers. Also, the present invention relates to a three-dimensional cell culture method comprising: a first step of dispensing cell suspension having cells suspended in culture media above a level including the holes in the culture space formed in the three-dimensional cell culture system; and a second step of removing excessive culture liquid, so that cells can be trapped in the holes. Also, the present invention relates to a drug screening system including the three-dimensional cell culture system, and further including one or more wells for containing drugs which have been formed deeper than the holes and in width including all the holes in parallel to the holes arranged in the rows and columns of the cell culture system outside the cell culture system. Also the present invention relates to a drug screening method comprising: a first step of dispensing cell suspension having cells suspended in culture media above a level including the holes in the culture space formed in the drug screening system; a second step of removing excessive culture liquid so that cells can be trapped in the holes; a third step of inputting a drug into the wells for containing a drug and transmitting the drug to the cell culture holes by diffusing the drug through the pores of the porous polymers; and a fourth step of detecting the reaction of the cells in contact with the drug transmitted to the cell culture holes.
