Provided are a composition for enhancing immunity, containing a milk protein with a high content of organic zinc or organic calcium as an active ingredient, and a method for preparing the same. The milk protein with a high content of organic zinc or organic calcium increases the bioavailability of minerals, wherein the milk protein with a high content of organic zinc or organic calcium is prepared by mixing mixed powder, which is prepared by mixing whey powder and water-soluble zinc or calcium powder, with water and then heating the same. In addition, the composition has immune enhancing activities by: increasing the number of immune cells such as macrophages, neutrophils, monocytes, etc., which primarily defend against pathogenic microorganisms, and natural killer (NK) cells in the blood; increasing the number of CD4 positive helper T cells (T_H cells) and CD8 positive cytotoxic T cells (T_C cells) in blood or lymph nodes; and increasing the secretion of tumor necrosis factor- (TNF-) and interleukin-1 (IL-1), the immune-related cytokines secreted from activated macrophages, when the composition is orally administrated, thereby being able to be used as an active ingredient of a food composition or a pharmaceutical composition for enhancing immunity.
