The present invention relates to a H1N1-based avian influenza virus highly replicative in an embryonated egg and avirulent in a mouse, a composition for manufacturing the virus, a manufacturing method of the virus, an avian influenza virus vaccine comprising the virus as an effective component, and a composition comprising an antiserum against the virus for diagnosing an avian influenza virus. The composition for manufacturing a H1N1-based avian influenza virus highly replicative in an embryonated egg and avirulent in a mouse comprises: a polynucleotide coding hemagglutinin (HA), and a polynucleotide coding neuraminidase of a mutant of a H5N1 virus highly replicative in an embryonated egg; a polynucleotide coding polymerase basic protein 2 (PB2) of a low pathogenic avian influenza; a polynucleotide coding a polymerase basic protein 1 (PB1), a polynucleotide coding polymerase subunit A (PA), a polynucleotide coding nucleoprotein (NP), a polynucleotide coding a matrix protein (M), and a polynucleotide coding nonstructural protein (NS) of a H1N1-based avian influenza virus.;COPYRIGHT KIPO 2016
