The present invention relates to vinegar of rhizome of Imperata cylindrical Beauvois and a manufacturing method thereof. Specifically, the manufacturing method comprises the steps of: 1) applying sugar to a rhizome extract of Imperata cylindrical Beauvois to manufacture a raw material having 10-20 of Brix; 2) inoculating yeast bacteria in the raw material to ferment the same with alcohol; and 3) inoculating acetobacter in the alcohol fermented product manufactured in step 2) to ferment the same with the acetobacter. According to the present invention, the vinegar of rhizome of Imperata cylindrical Beauvois manufactured by the manufacturing method has abundant essential amino acids, and has excellent taste and smell, thereby being broadly used as vinegar or a beverage composition.
