PROBLEM TO BE SOLVED: To provide a skin penetrability assessment method that captures a structural change in a skin horny layer intercellular lipid by administration of a test substance, which cannot be measured by typical skin penetrability test methods, by an X-ray diffraction measurement, and also enables a measurement of skin penetrability of the test substance in a molecule level.SOLUTION: A technique is found that is adapted to: perform an X-ray diffraction measurement of a skin horny layer having a test substance administered and a skin horny layer not having the test substance administered as a target; detect a significant difference in a peak area depending upon presence/absence of the test substance administration from a peak deriving from a lamellar structure of a skin horny layer intercellular lipid and a packing structure or a peak deriving from the test substance from a small angle area of a diffraction profile obtained from the X-ray diffraction measurement and a wide-angle area thereof; and thereby asses penetrability to the skin horny layer of the test substance. Accordingly, the present invention is completed.
