The present invention relates to an improved composition of mesenchymal stromal cells (MSC) and its preparation. The present invention relates to a new strategy for isolating MSC from bone marrow mononuclear cells (BM-MNC) by combining BM-MNC from unrelated (third party) bone marrow donors. The MSC composition, manufactured according to the method of the present invention, is characterized by a stable proliferative capacity and an increased immunosuppressive potential compared with the MSC composition from individual donors or a pool of individual MSCs obtained from several donors. The MSC obtained in accordance with the present invention is particularly useful for medical applications, such as the treatment of graft versus host disease (GvHD) in recipients with hematopoietic stem cell transplants in patients with autoimmune disorders, and as cell therapy in regenerative medicine.
