The present invention reports production of bacterial cellulose in natural media by potent cellulose producing bacterial isolate, Komagataeibacter hansenii RV39. The organism was isolated from rotten sweet lime fruit and is deposited at MTCC, IMTECH, Chandigarh under the deposition number MTCC 25039. Characterization of the produced bacterial cellulose was done using Scanning electron microscope (SEM), Fourier transform infrared spectroscopy ..(FTIR), X-ray diffraction (XRD), Nuclear magnetic resonance (NMR), Thermogravimetric analysis (TGA) and Differential scanning calprimetry (DSC). Further a process for cellulose production was developed optimizing the physical and nutritional parameters. The present invention also recites the production of bacterial cellulose from different natural source media. The cellulose yield from ripped water melon and tomato juice was found to be 12.1g/l and 13.31g/l which is almost double as compared to yield in standard medium (7.2g/l). Further, the invention describes utilization of rind part of watermelon, jack fruit, cabbage, pineapple and carrot for the production of bacteria cellulose. Yield obtained from pineapple and watermelon rind extract was 4.5g/l to 5.3 g/1 which was lesser than ripped tomato and watermelon but it is highly significant and cost effective cheaper source.
