摘要

ABSTRACT The present invention relates with a new strategy forefficient trans-differentiation of human hepatic progenitor cells (hHPCs)into insulin producing cells (InPCs) on 3D-nanostructured TiO2 sheetsto provide long-term cell survival and function. The another embodiment of this invention describes about the applicability of cellularized nanostructured TiO2implants suitable for ectopic transplantation in diabetic mice where it elicits very limited immunological response and shows high degree of in vitro and in vivo biocompatibility. Further the applicability of this cellularized implant was investigated in diabetic mice. The preliminary investigation revealed that the TiO2 sheets on conducting substrates provided better support for in-vitro trans-differentiation and expansion of human hepatic progenitors into InPCs. Towards the application the major finding revealed recovery of blood insulin level and long-term glycemic control in hyperglycemic mice without immunosuppressant. This invention relies to step forward for resolving the problem of unavailability of quality donors and continuous need of immune suppression during whole pancreas and/or cadaveric islets transplantation which areone of the conclusive methods for treatment of uncontrolled diabetes.
