The present invention refers to a novel in vitro culture composition free of serum for obtaining erythrocytes parasiteinfested with Babesia spp. and the production of an attenuated living vaccine reactant. The living in vitro culture composition of the present invention comprises a serum-free basal medium combined with at least a supplement for basal medium that contains insulin, transferrin, selenite, albumin, linoleic acid and oleic acid; and the addition of putrescine, preferably for the obtention of erythrocytes parsiteinfested with babesia bovis and/or Babesia bigemina. The present invention also refers to the obtention of soluble antigens and/or particulate antigens obtained from the serum-free in vitro culture composition, said antigen being used for serological tests with reduction of nonspecificity upon performing ELISA and/or indirect immunofluorescence readings.
