Provided is a method for using CRISPR-Cas9 specificity to knock out a swine leukocyte antigen 2 (SLA-2) gene, and an sgRNA used for specifically targeting the SLA-2 gene. The target sequence of the sgRNA for specifically targeting the SLA-2 gene conforms to 5'-N(20)NGG-3' sequence rules, N(20) representing 20 consecutive bases and N representing A or T or C or G; the target sequence in the SLA-2 gene is unique and is located at the 5 exon coding regions, or the junction with the adjacent introns, at the N end of the SLA-2 gene.
