Provided are a method for pretreating a high-concentration organic matter sample and a method for measuring heavy metal content. The pretreatment method being a separating digestion method, and comprises: an acid or acid solution is added to a sample uniformly, at this time the volume of the acid or acid solution is 2-100 times that of the sample, and heating is performed to boiling, with the heating temperature controlled to prevent dryout, resulting in sample carbonization; by means of the temperature and process of heating and boiling water and the acid and also the temperature and process of generating acid smoke during the acid heating and boiling, a solid or liquid organic matter with a higher molecular weight in the blood sample being converted into a liquid organic matter remaining in the pretreated solution, and after the completion of the heating, the solution is slightly cooled, and boiling continues after the addition of the acid solution, at this time the volume of the acid solution is 2-100 times that of the sample; and then the temperature is reduced to room temperature, resulting in grease solidification, and after solid-liquid separation or oil-water separation, a digested solution is obtained. Compared with the prior art, such a method can substantially reduce the concentration of an organic matter with a higher molecular weight in a sample, and improve the accuracy of the concentration measurement.
