The present invention relates to a dark field lighting-based polarization-sensitive optical coherence tomography (PS-OCT) system and a method thereof. The method comprises: a separation step of separating light generated from a light source into reference light and irradiation emission light; a polarization step of polarizing the irradiation light; an irradiation step of irradiating the polarized irradiation light on a sample in a Bessel beam type; a detection step of detecting detection light which is irradiated on the sample; a calculation step of converting the reference light and the detection light into digital signals, and calculating polarization characteristics of a tissue of the sample; and a display step of displaying an image according to the polarization characteristics. According to the present invention, since a collection of surficial reflective signals can be minimized due to an application of a dark field lighting type, a correct image can be acquired, thereby extended application and practicality in clinical application fields can be increased.;COPYRIGHT KIPO 2017
