The present invention relates to a method of manufacturing a mixed extract of Inonotus obliquus, Phellinus linteus, and Sparassis crispa, by producing complex mycelia by mixing mycelia of Inonotus obliquus, Phellinus linteus, and Sparassis crispa at a predetermined weight ratio, and extracting the complex mycelia in a double boiler at a constant temperature and a high pressure or vacuum environment; and to the extract manufactured therefrom. The extract contains a lot of bioactive substances of the mushrooms, particularly beta-glucan which has an excellent anticancer effect. According to the present invention, the method comprises the followings steps: cultivating Inonotus obliquus, Phellinus linteus, and Sparassis crispa; washing each mushroom with water to remove foreign materials; drying each mushroom to have a moisture content of 15% or less; finely cutting each mushroom to prepare pulverized mushrooms; mixing the pulverized mushrooms at a predetermined weight ratio to obtain a mushroom mixture, and inserting the mushroom mixture into a high pressure tank; and extracting an extract from the high pressure tank at a temperature of 55 to 65C and a tank pressure of 1.5 to 3.0 atm for 2 to 4 days.
