The present invention relates to a hyaluronic acid derivative hydrogel manufactured by cross-linking using an akinetic click chemistry reaction which does not need a copper catalyst having cytotoxicity, to a preparation method thereof and an in vivo gelation method using the same. According to the present invention, through cross-linking using an akinetic click chemistry reaction which does not need a copper catalyst having cytotoxicity, a hydrogel can be formed in a desired position in a living body. The formed hyaluronic acid derivative hydrogel maintains long-term stability in human bodies and has excellent biocompatibility, thereby being used as a filler and a carrier for transferring a cell, a gene and a protein to improve wrinkles.;COPYRIGHT KIPO 2017
