The present invention provides designer nucleic acid constructs and polypeptides that can be used as therapeutic vaccines against HPV 16. These polypeptides essentially contain all possible T-cell epitopes of the HPV 16 tumor proteins E6 and E7, but nevertheless contain a minimal number of undesired neo-epitopes, as well as fragments of rearranged E6 and E7 proteins (Compared to wt E6 and E7), to a degree of non-detectable transformation activity. The invention provides a nucleic acid molecule encoding a polypeptide comprising the sequence set forth in SEQ ID NO: 1. In certain embodiments, the encoded polypeptide further comprises at least one epitope of a human papilloma virus (HPV) E2 protein, e. G. HPV 16 E2 protein.
