The present invention relates to a betaglucan-peptide complex, a preparation method of the same, and a pharmaceutical composition for preventing or treating inflammatory diseases comprising the same as an active ingredient. Specifically, shizophyllan (SPG) which is beta-1,3-glucan, is covalently bound to SH3 peptide originated from c-Src to form SPG-c-Src-SH3 peptide complex, and the pharmaceutical composition comprises the complex as an active ingredient for preventing or treating inflammatory diseases such as sepsis and colitis. The betaglucan-peptide complex according to the present invention significantly reduces the production of active oxygen (ROS) and inflammatory cytokines by targeting p47phox, and thus can be beneficially used as a pharmaceutical composition for preventing or treating ROS-related life-threatening inflammatory diseases such as sepsis and colitis. Further, the betaglucan-peptide complex according to the present invention can be beneficially used in the development of novel peptide-based drugs targeting p47phox.
