The present invention relates to a method for preparing induced pluripotent stem (iPS) cells. The iPS cell preparing method includes the steps of: (a) culturing human-derived somatic cells applied with genes encoding a reprogramming inducer in a culture medium including one or more selected reprogramming-enhancing agents selected from a group including protein kinase C inhibitors, histone deacetylase inhibitors, and osteogenic protein pathway inhibitors; and (b) separating embryonic stem cell-like colonies from the cultured product obtained from step (a). According to the present invention, the method enables preparation of the iPS cells with high efficiency or enable the preparation of the iPS cells even under xenopathogen-free and feeder cell-free conditions.
