Provided is a method for targeting cells for depletion, including but not limited to tumor cells, in a therapeutic regimen that includes contacting target cells with a combination of agents that modulate immunomodulatory signaling. . Immunomodulatory agents include (i) agents that block CD47 activity, and (ii) agents that stimulate immune costimulatory molecules such as CD137. The therapeutic regimen may further include an agent that specifically binds to the target cell, eg, an antibody or biologically active fragment or derivative thereof. Target cell depletion levels are high for treatment regimes where a single immunomodulator is used, and the effect is synergistic for a single treatment regime where a single immunomodulator is used obtain.
