PROBLEM TO BE SOLVED: To provide a method for specifically cleaving Cα-C bond and/or side chain of peptide main chain of protein and peptide, and a method for determining the amino acid sequence of proteins and peptides.SOLUTION: A method for specifically cleaving Cα-C bond and/or side chain bond of peptide main chain includes a step of irradiating the protein or peptide with laser light in the presence of hydroxynitrobenzoic acid selected from a group consisting of 3-hydroxy-2-nitrobenzoic acid, 4-hydroxy-3-nitrobenzoic acid, 5-hydroxy-2-nitrobenzoic acid, 3-hydroxy-5-nitrobenzoic acid, and 4-hydroxy-2-nitrobenzoic acid. A method for determining amino acid sequence of protein or peptide includes the steps of: irradiating the protein or peptide with laser light in the presence of the above specific hydroxynitrobenzoic acid; specifically cleaving the Cα-C bond and/or the side chain bond of the peptide main chain; and analyzing the generated fragment ions by mass spectrometry.SELECTED DRAWING: Figure 2
