A genetically engineered payload delivery system includes a target cell binding unit covalently attached to a pore-forming unit and a payload portion adapted with a region that can be non-covalently attached to the pore-forming unit. The pore-forming unit is from a specific serosubtype of Clostridial toxin, while the payload region is from another serosubtype of Clostridial toxin. The chimeric protein-based compositions of the present disclosure can deliver payload specifically to neurons.
