RUFINAMIDE AND DERIVATIVES AND THEIR USE IN MODULATING THE GATING PROCESS OF HUMAN VOLTAGE-GATED SODIUM CHANNELS

The present invention provides compounds of formula I or a salt, solvate, or stereoisomer thereof, wherein X is H, or an electron withdrawing group such as a halogen, NH₂, NO₂, SO₂, CN, or a C₁-C₆ alkyl group; Alk is C₁-C₃ alkyl; R₁ is H, C₁-C₆ alkyl, which may be substituted with OH, NH₂, acyl, sulfonyl, and cyano groups; and R₂, is C₁-C₆ alkyl, which may be substituted with OH, NH₂, acyl, sulfonyl, and cyano groups. Pharmaceutical compositions comprising these compounds and/or rufinamide are also provided. Methods for prevention and treatment of epilepsy disorders such as Lennox-Gastaut Syndrome (LGS), and modulation of voltage-gated sodium (Nav) channels are Nav1.1 channels by administration of these compounds and/or rufinamide are also provided.
