The present invention relates to a protein designed and generated by genetic engineering by a synthetic gene coded from the 27 kDa protein. The 27 kDa protein forms a three-dimensional structure similar to the nucleotide-binding domain or N-domain (amino acid residues 360 to 601) of Ca2 + ATPase, characterized by intrinsic fluorescence highly sensitive to the union of nucleotides. The high sensitivity in the fluorescence of the 27 kDa protein allows assays of binding/interaction of molecules with affinity to the nucleotide binding site. In the case of fluorescent ligands, it realizing FRET assays and measuring molecular distances.
