A method for the production of reactive human donor regulatory T cells (Treg), comprising: a) co-culturing CD19 + B cells from a 5-year donor with irradiated CD40L + human leukemia feeding cells under conditions effective to produce stimulated B lymphocytes ( sBc); b) co-culturing CD4 +, CD25 +, CD127- / l or T cells from a human recipient with said sBc under conditions effective to selectively expand reactive human donor regulatory T cells (Treg); and c) restimulating said donor reactive Treg by crosslinking CD3 and CD28 of said donor reactive Treg under conditions effective to produce donor reactive Treg which are CD4 +, Helios + and Foxp3 +; where the donor is HLA mismatch relative to the human recipient.
