Adoptive transfer of CD8 + T cell clones derived from central memory cells

A method for obtaining a primate CD8 + cytotoxic T lymphocyte (CTL) preparation, useful for adoptive immunotherapy comprising the steps of: (a) enriching a population of T lymphocytes obtained from a donor primate for a subpopulation of memory T lymphocytes central CD62L + CD8 + (TCM), to thus generate an enriched composition of TCM that is enriched with CDM lymphocytes CD62L + and depleted of memory T lymphocytes effector CD62L- (TEM); and (b) expanding the cells of the enriched TCM composition in vitro, optionally in the presence of interleukin (IL) 15 to produce a CTL-enriched CTL preparation derived from TCM, the CTL preparation comprises effector T cells (TE) , wherein TE cells have a decrease in CD62L expression compared to the enriched TCM composition of step (a).
