A method for analyzing a population of seeds, the method comprising: exposing the endosperm of the seed of individual seeds in a population of seeds while the germination viability of the seed is retained, drench viable seed in an alkaline solution thereby forming a soaking solution seed of at least one of the individual seeds in the seed population and analyzing the solution soaking seeds for the presence or absence of one or more traits of interest, wherein the isolated DNA in the soaking solution of seed is evaluated for one or more traits of interest comprising a genetic marker, a polymorphism of a single nucleotide, one simple sequence repeat, a haplotype.
